purpose And Objectives: A Major Objective Of The National Institute On Aging (nia) Is To Identify And Characterize Molecular Derangements That Occur Early In The Alzheimer’s Disease (ad) Process, Both In Brain And In Blood. The Lipid Peroxidation Unit Places A Particular Emphasis On Identifying Derangments In Lipid Metabolism-related Pathways And Markers (such As Apoe). To Achieve These Objectives, Nia Requires Rapidly Autopsied Specimens From Key Regions That Are Involved In The Earliest Stages Of Ad (pons, Entorhinal Cortex, Hippocampus) From A Cohort That Spans The Full Spectrum Of Ad. Most Of Nia’s Lipid Metabolism Related Assays And Ihc Cytoarchitectural Markers Require Formalin-fixed Paraffin Embedded (ffpe) Specimens. Others Require Frozen Specimens. Therefore, Nia Requires Both Types Of Specimens, Preferably From The Same Cohort Of Autopsied Participants.
project Requirements: Markers In Tissue Specimens Begin To Degrade Rapidly After Death. Nia Requires Tissue Specimens From Rapidly-autopsied Participants, With An Average Post-mortem Interval Of 3 Hours. Nia Also Requires Tissues From The Full Spectrum Of Alzheimer’s Disease And From Regions Involved In The Earliest Stages Of Disease. Each Of These Elements Are Essential For Testing Hypotheses.
nia Requires Twenty (20) Sections From Each Of 30 Cases Spanning The Clinicopatholgical Spectrum Of Ad From Each Of The 3 Regions (1,800 Total Sections), Plus Corresponding Frozen Specimens And Plasma, And Positive And Negative Control Brain Blocks.
